What is Selank?
Selank is a heptapeptide developed in Russia. It is a synthetic analog of the peptide tuftsin. Research confirms the importance of tuftsin and its derivatives, as well as analogs such as selank. Most of these substances have demonstrated antitumor, anti-inflammatory, and antibacterial activities. Tuftsin can also be used in the preparation of fusion proteins for cancer therapy.
Furthermore, some researchers have made significant progress and observed encouraging effects of Selank on morphological parameters in cases of chronic stress. Clinical studies have also demonstrated that Selank has an anxiolytic effect comparable to that of benzodiazepines. The molecular mechanism of action of Selank may be related to its ability to modulate the functioning of the GABAergic system. Moreover, studies have confirmed the hypothesis that this peptide may affect the interaction of GABA with GABAA receptors. Selank has also been shown to influence serotonin metabolism in various ways.
Advantages of Selank research
Molecular aspects of the anxiolytic effect of Selank
Researchers have suggested that selank acts as a positive allosteric modulator of [3H]GABA binding. The combined action of selank and other benzodiazepines also regulates [3H]GABA binding activity in a specific, non-cumulative manner, and differently for each peptide.
Selank can block the modulatory activity of diazepam and olanzapine; however, their peptide-binding sites and localization differ, although there may be some partial overlap. Studies have hypothesized that one of the anxiolytic molecular mechanisms of Selank may be associated with allosteric modulation of GABA receptors, which is concentration-dependent and receptor subtype-selective.
Selank in the regulation of adaptive behavior in stressful situations
White male laboratory rats, pre-sorted according to their emotional reactivity, were used to study the effects of Selank on the behavioral manifestations of emotional stress induced by a conflict situation. The peptides generally showed positive emotional effects and an anti-stress action. Significant individual physiological effects were observed, linked to the molecular structure of the peptides studied. The results of this study demonstrate the potential for the synthesis of peptide compounds with predictable pharmacological actions and good tolerability, for large-scale use.
The role of Selank in the immune system
Immunomodulation is an important therapeutic method. It involves stimulating or inhibiting the immune system using various substances called immunomodulators. These compounds are designed to regulate the body’s immune response to infections of various etiologies. Tuftsin and some of its analogs are examples.
According to a new study, selanke induces a transcriptomic response in the hippocampus and spleen cells of rats. It could play a role in regulating inflammatory processes in the body.
Several studies have examined the effect of Selank and two of its fragments on the expression of genes involved in inflammatory processes. Scientists analyzed the expression of these genes in the spleens of mice 6 and 24 hours after a single intraperitoneal administration of Selank. They found changes in the expression of nearly half of the genes analyzed. The Bcl6 gene, which plays a central role in the formation and development of the immune system, showed significant variations in its expression level.
Furthermore, research has shown that Selank induces several alterations in the expression of genes playing a major role in inflammation. This complex and significant biological effect of Selank could be partly determined by its systemic impact on gene expression.
Selank’s effect on memory enhancement research
The effect of Selank on learning and memory processes has been studied experimentally in rats. This study also showed that Selank can influence serotonin metabolism.
Selank or a saline solution was administered, and the conditioned feeding reflex was further developed 30 minutes later. Retention was tested 24 hours, 7 days, and 30 days after Selank administration. A single dose of Selank activated serotonin metabolism in the hypothalamus and caudal brainstem cells for 30 minutes to 2 hours.
Scientists have concluded that Selank induces and strengthens the stability of memory traces. Based on this knowledge, administered Selank can improve memory processes. This activity of Selank is likely due to its effect on serotonin levels and its metabolite, particularly in the brain.
Résumé
According to various studies and medical researchers, Selank, heptapeptide and synthetic tuftsin analogues have shown potential benefits as immunomodulators, as well as in their action on helper T cells and their regulation.
Human studies have shown that Selank could potentially improve memory and mood. This peptide may also have a positive impact on emotional well-being and reduce stress.
